This MAb recognizes cytokeratin 4, 5, 6, 8, 10, 13, and 18. This is a broad-spectrum antibody, which has been reported to differentiate epithelial tumors from non-epithelial tumors. Recognizes an N-glycosylated glycoprotein of 120 kDa with intra-chain disulfide bonds, identified as CD50 or ICAM-3 (WS: IV & V). Its epitope localizes in the D2 extracellular domain and is resistant to neuraminidase and proteases. CD50 is the major ligand for LFA-1 (CD11a/CD18) and may have signalling role to increase adhesion. It is expressed on thymocytes and T lymphocytes and is resistant to treatment with phosphatidylinositol (PI) phospholipase C. This MAb inhibits primary mixed lymphocyte culture (MLC) but not secondary MLC, cytotoxicity or proliferation induced by mitogens. It blocks binding of NK1-L16 stimulated T cells to L cells expressing CD50. CD10 (also known as Common Acute Lymphocytic Leukemia Antigen (CALLA)) is a type II transmembrane protein. It is a cell surface enzyme with neutral metalloendopeptidase activity, which inactivates a variety of biologically active peptides. CD10 is expressed on the cells of lymphoblastic, Burkitt's, and follicular germinal center lymphomas, and on cells from patients with chronic myelocytic leukemia (CML). Reacts with human CD48, a 45 kDa glycosyl phophatidyl-inositol (GPI)-anchored cell surface protein. CD48 is strongly expressed on lymphocytes and monocytes and weakly on granulocytes but is absent on platelets, fibroblasts, epithelium and endothelium. Recognizes a protein of 36 kDa, identified as cyclin D1. Cyclin D1, one of the key cell cycle regulators, is a putative proto-oncogene overexpressed in a wide variety of human neoplasms. This antibody neutralizes the activity of cyclin D1 in vivo. About 60% of mantle cell lymphomas (MCL) contain a t(11; 14)(q13; q32) translocation resulting in over-expression of cyclin D1. This antibody is useful in identifying mantle cell lymphomas (cyclin D1 positive) from CLL/SLL and follicular lymphomas (cyclin D1 negative). About 40% of breast carcinomas are positive for Cyclin D1. Recognizes a protein of 205 kDa-220 kDa, identified as CD45RA (Workshop III). CD45RA is isoforms of the human leukocyte common antigen (CD45). Human CD45 contains three exons which encode peptide segments designated A, B and C, respectively. The differential splicing of the exons generates at least five isoforms, ABC, AB, BC, B and O. This antibody reacts with ABC and BC isoforms. CD45RA is expressed on 40-50% of peripheral CD4 T-cells, 50% of peripheral CD8 T-cells, B-cells, and leukemic B-cell lines. T-cells expressing CD45RA are naive or virgin T-cells. T-cells expressing CD45RO are memory T-cells. CD45RA and CD45RO define complementary, predominantly non-overlapping populations of resting peripheral T-cells. This MAb is useful in study on the subpopulation of CD4 or CD8 T-cells. This MAb reacts with an extracellular domain (close to transmembrane) of CD56/NCAM. Three isoforms of neural cell adhesion molecule (NCAM) are produced by differential splicing of the RNA transcript from a single gene. The 135 kDa isoform is the basic molecule, which is glycosylated or sialylated to produce the mature species. Anti-CD56 recognizes two proteins of the neural cell adhesion molecule, the basic molecule expressed on most neuroectodermally derived tissues and neoplasms (e.g. retinoblastoma, medulloblastomas, astrocytomas, neuroblastomas, and small cell carcinomas). It is also expressed on some mesodermally derived tumors (rhabdomyosarcoma). This antibody recognizes proteins of 80-200 kDa, identified as different members of CEA family. CEA is synthesized during development in the fetal gut and is re-expressed in increased amounts in intestinal carcinomas and several other tumors. This MAb does not react with nonspecific cross-reacting antigen (NCA) and with human polymorphonuclear leucocytes. It shows no reaction with a variety of normal tissues and is suitable for staining of formalin/paraffin tissues. CEA is not found in benign glands, stroma, or malignant prostatic cells. Antibody to CEA is useful in detecting early foci of gastric carcinoma and in distinguishing pulmonary adenocarcinomas (60-70% are CEA positive) from pleural mesotheliomas (rarely or weakly CEA positive). This MAb reacts with a wide variety of simple epithelia. It does not react with stratified squamous epithelia. It reacts with epithelial tumors of the gastrointestinal tract, lung, breast, pancreas, ovary, and thyroid. Cytokeratin 18, which belongs to the type A (acidic) subfamily of low molecular weight keratins, exists in combination with cytokeratin 8. It was reported that tissues from gastrointestinal tract are positive for both cytokeratin 8 and 18 but do not contain cytokeratin 14. Biotium Chromogranin A(CGA/414), CF488A conjugate, 0.1mg/mL
Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms.
